Effects of HO-1 modified BMMSC on the immunological function of small bowel transplantation in rats

HO-1 修饰的 BMMSC 对大鼠小肠移植免疫功能的影响

Abstract: Objective To investigate whether heme oxygenase-1(HO-1) could enhance the immunosuppressive effects of bone marrow mesenchymal stem cells(BMMSC)on the rejection of transplanted small bowel allograft in rats. Methods The rejection model of ectopic small bowel transplantation in rats was established,then BMMSC or HO-1/BMMSC were transfused via the dorsal penis vein. The histopathological changes of the transplanted small bowel and the change of regulatory T cell(Tregs),natural killer cell(NK)and cytokines after transplantation were observed in different groups. Results The acute rejection model rats after small bowel transplantation showed mild to moderate on post operation day 5(POD5)and severe acute rejection on POD7. BMMSC showed strong protective effects on rejection within the first 7 days after transplantation,anda downward trend since POD10. While HO-1/BMMSC showed stronger effects than BMMSC,which provided enhanced protective effects on the transplanted small bowel. In addition,the activity of natural killer(NK) cell decreased significantly,the levels of regulatory T cells(Tregs),interleukin-10(IL-10)and transforming growth factor-β(TGF-β) increased significantly, and the levels of interleukin-2(IL-2), interleukin-6 (IL-6),interleukin-17(IL-17),interleukin-23(IL-23),tumor necrosis factor -α(TNF-α)and interferon-γ (IFN-γ)decreased significantly in the HO-1/BMMSC group, when compared with the BMMSC group. Conclusion HO-1/BMMSC showed better immunosuppressive effects after transplantation. HO-1 could enhance the effects of BMMSC inhibiting the immune response of acute rejection in ectopic small bowel transplantation model of rats.

摘要: 目的 探讨在小肠移植排斥反应中血红素加氧酶 -1(HO-1)是否能增强骨髓间充质干细胞 (BMMSC)对移植免疫功能的影响。方法 建立异位大鼠小肠移植排斥反应模型,实验分为生理盐水组(NS)、 BMMSC 组和 HO-1/BMMSC 组,经阴茎背静脉给予输注 BMMSC 和 HO-1/BMMSC,观察术后各组移植小肠 组织病理、调节 T 细胞(Tregs)、自然杀伤细胞(NK)及细胞因子的改变。结果 小肠移植急性排斥反 应模型组术后 5 天发生轻至中度的急性排斥反应,术后 7 天发生中至重度的急性排斥反应,术后第 10 天 发生重度的急性排斥反应。HO-1/BMMSC 处理组与 BMMSC 处理组相比,显著提高了 BMMSC 保护移植 小肠的作用 ;并且 NK 细胞活性显著下降而 Tregs 水平显著升高 ;细胞因子白细胞介素 -10(IL-10)、转 化生长因子 -β(TGF-β)显著上升而白细胞介素 -2(IL-2)、白细胞介素 - 6(IL-6)、白细胞介素 -17 (IL-17)、白细胞介素 -23(IL-23)、肿瘤坏死因子 -α(TNF-α)及 γ- 干扰素(IFN-γ)则显著降低。 结论 HO-1/BMMSC 能够更好地发挥移植免疫抑制作用,HO-1 可增强 BMMSC 抑制小肠移植急性排斥 反应大鼠体内免疫功能的作用。
